Yes it is possible. Are the two computers connected in a network together already and do you have a router? If you don't have a router then you could get one and have it upstairs connected to the desktop and if your laptop doesn't have one already, get a wireless card for your laptop. Once the desktop and laptop are in the same network you can do stuff like create shared folders and printer sharing. Enable printer sharing on the laptop and then the desktop should be able to print from the laptop when it is turned on.
