AGP or PCI - Is there any difference?

AGP is a faster interface than PCI, also AGP cards are cheaper than a PCI equivalent due to them being in greater supply. PCI Express however, is faster than AGP but right now there is no tangible benefit

Isn't PCI Express just AGP 16X fancified up?

there is no AGP 16x so it'd be hard to fancy it up. basically PCIe is just a full duplex bus, then you can add lanes to increase the speed
