Can I boot Lucid Puppy from hard drive as my main OS?

You have to install it on a hard drive partition. You should be able to create a partition using that live CD.
 
what is the swap partition used for? I did create a swap partition and only gave it 250mb. Is it alright?
I believe the ext3 partition is used for puppy installation...?

swap is linux's equivelent of windows' virtual memory. its what the system uses when RAM is running low. it uses it as a temporary RAM storage, and it acts by swapping modules of RAM to and from swap space. (source)

The size of the swap is best to be the size of your ram. eg If you have 2gb of ram, get a swap partition that's 2gb
 
So I will make a swap partition of 64mb (actual ram of my old toshiba laptop) and see how it goes.
