Can you unmount a game and still play it?

I got Skyrim off of piratebay (I did purchase it for Xbox 360, at least), and I was wondering if I can get rid of the iso file on my computer and still play the game. Obviously I need to unmount the game before I can delete the iso file, but will unmounting it and deleting that file cause Skyrim to be unplayable? I installed skyrim and put in the crack already, and the game plays well. I'm just a little bit of a neat freak when it comes to my computer and unnecessary files.
