Is it better to have (2) 512mb sticks of ram, or (1) 256mb and (1) 1gb?

Is there any problems with having it so lopsided in the slots? (256mb and 1gb)

Dual channel will work better. (the two in a pack)

Having different amounts is slots won't cause any problems.
