I'm about to start a new build for the first time.
I want to build it outside of the box to make sure that it's running OK. Is it necessary for me to have the heatsink attached on my initial start-up before I actually make the build in the case?

Yes. No matter what you ALWAYS need to have a heatsink on the CPU when you power it up.
 
Make sure you are clear of ESD.. Working outside of the box is a little worse than working inside with a grounded Motherboard.
