Every motherboard has pins on which you must connect the USB wires.If you connect the wires on those pins the wrong way than USB port logically won't work.For example my motherboard has 2 USB connectors and each has 9 pins,but only 5 of those 9 pins is used to make a USB connection while other 4 remains intact.Same applies for other USB connector of 9 pins.
So...how many pins your USB connector(s) have?
Because according to that we can know how you must connect your USB in order to make your devices attached to that USB to work.
For example this is what I have used to add one more USB entrance on my computer:
