Back in November last year I imaged my C: drive and keep the image on D:
Now today by mistake I included the image file when defragging D: drive and it spent the best part of 4 minutes moving the clusters of the image .
My question is will this have any affect if I decide one day to use that image because when the image was made it was fragmented and now the image is de-fragmented.

It shouldn't. I think it will be fine.

If you wanted to test it:

1) Make a new fresh image today.
2) Revert to the old (questionable) image.
3) If it works fine (which it should), revert again to the image you created today.
4) Delete today's image and save the old one for when you need it.
