Only Ram Pc

hello,
Question ,
Can A fully operating system + programs + pc setting etc be loaded from a
bootable DVD, usb key to eg. 16 Gb of phisical RAM ( not iRAM or HyperOs )
remove the disk or key and use the pc normaly ?
As OS it means full Vista 32 or 64 , not small like puppy linux etc
hope some can advice on

I understand about shut it down, but can I still do : better explained > I install an Os, all my programms, etc from some kind of removable media ( not matter if take 10 , 30 minutes to load) remove the media > is it possible ? and so Has the pc all data necessary to work,? I know windows 32 bit eg ,does not address more than 3/4 Gb can i some how configure 16/32 RAM as HD ?
