Recently I have been modding a controller and accidently removed the tin underneath the button connector grid and need to connect wire to this area, I am wondering if I need that small layer of tin on the PCB or if just soldering the wire into where the tin was is good enough.

Do you mean the pointy thing coming out of the bottom of the PCB? I don't think that's too important, try running it without it.
