My advice would first be to fire up the game and play it until it hits a slowdown period you described. Exit out of the game and start the task manager (alt+ctrl+del) and select the performance tab. In the box that is labeled "commit charge" look for the line that says "peak" if the amount of memory used during the peak time is much more than the amount of physical memory you have in your computer you will experience a rather long lag during play when the game is trying to load something. The only fix there is to add more RAM to the computer.

Any number of things could cause suddenly use more memory than before. Say after installing Service Pack 2 or installing a new program that is running in the background, some updates to antivirus software can cause it to use more memory than before. ( I would suggest shutting off your antivirus during game play)

You might also try updating your antivirus and running a full system scan to see if there is a virus thread using up some of your memory.
