USB flash as a HD

If one is installing a program and the program asks where to install the files. Is it possible to use a flash drive (16gb) as the storage. Will the program find the files or do they need to be on the main HD (local C) ? Also does a 64bit differ on program installs than a 32bit because of the (2) program files folders?
 
I figure it should be, When you download save it to your USB, in the Save window, go to Computer, then select your USB from that screen.
