External Enclosure vs. External Hard Drive

So I am looking to get a hard drive of some type that would be large enough to back up my 320gb internal hard drive for a reasonable price.

I have a few questions about how to go about doing this:
-I know it's possible to back up my computer on an external hard drive (I've done it before), but is it possible to do it using an internal hard drive in an external enclosure?
-Is there a difference between the data transfer rate between the two?
-Is one more reliable in any way than the other would be?
-Which route would happen to be the most cost effective?

And, of course, does anyone have a recommendation that they would make?

A internal drive inside an external enclosure is the same as a external drive. I'd go with a internal drive because back-ups would be faster. SATA to external USB are slower, and I've had faster system restores with my internal SATA back up drive. So I'd go with a internal drive.

Unless he uses eSATA. I use the eSATA with my drive and computer and it's very quick.
