There won't be any (real) speed benefits. However, the 2x4gb configuration will give you (if you have 4 slots in your motherboard) room to upgrade in the future. The counter argument being 4x2gb is usually cheaper.

I would go with 2x4gb.

2x4gb also = less draw from mb, easier on memory controller, and perhaps easier if oc'ing.
