DNS Servers Priority

If you set the DNS Servers on your Router and the DNS Servers on your local machine, which one of both will take priority?

Local settings. By manually specifying DNS servers, you're basically telling the PC to ignore the reference obtained from DHCP.

I just use the ipconfig /flushdns from time to time to get the best results.

As beers said, local settings take priority. Often times your consumer router will have DNS entries, and then tell DHCP connected clients to use your router as a DNS server. If you override that with your own entries, it will use those.
