What happens if I use portable hard drives for laptops to my desktop and vice versa, portable hard drives for desktop to my laptop? Any major damage?

Major damage? The only damage you could see would be any transfer of files between an external drive used between laptops and desktops that saw any virus infected files. The transfer would then spread the virus effecting Windows.

You could wear out a usb or eSata port from regular plugging and unplugging over a period of time. External drives are made for this type of use to begin with in order to allow moving data and files between laptops to desktops, laptops to laptops, desktops to desktops, etc. there.

External drives simply put are internal drives wrapped up in a self contained external casing. Just like the difference between 3 1/2" drives seen in desktops and 2 1/2" drives used on laptops/notebooks external drives can often be taken out of the external casing and simply be used inside the desktop's case or as a second drive when removing the cd/dvd drive in a portable.
