Moving major files from one drive to another

I moved itunes and myphotos from my C drive which is small to my E and D drive. Is it ok to just cut then paste the folder into the larger drive? Also for easy access, if it is in the E drive for example- do you just drag the folder onto the desktop then allowing if you need to upload photos or whatever you can just go onto desktop and find myphotos? How do you make a shortcut on the desktop for a large file in the E drive when the desktop is in the C drive?
 
You can make a shortcut for any Drive, Folder, or File by Rt Clicking on it and Clicking on Create Shortcut, or Send to Desktop (create Shortcut).

When moving the itunes, and Photo Folders you have have to point the Programs that use them to the new location from somewhere inside the programs preferences.

I am not familiar with those programs so don't know where you do it from
