Performance loss through USB port?

I was just wondering if there would be a performance difference in a game depending on where it was installed. The computer's HDD or a portable HDD connected through a usb port. Would the removable HDD slow the game or program down at all compared to it being on the computer's standard HDD?

I guess it might also depend on the speed of the HDD?

It most definitely will, hard drives connected via USB are significantly slower then ones attached via IDE/SATA. You will notice the biggest slowdown when it comes to game load times, and possibly during the game itself.
