I operate a Dell Dimension 4400, with two slots of RAM which currently house one 256mb stick and one 512mb of RAM for a total of 768mb. My motherboard can only hold two sticks. I was wondering if adding a 512mb stick in the place of the 256 will make much of a difference or not?

It won't make a really noticeable difference I'd say. If DDR was as cheap as DDR2 I'd say go for it, but unfortunately its $30 for 512MB vs. $10 for DDR2.
