Using a USB Floppy to update BIOS?

That will depend if you can boot from the drive with a prepared floppy with the flash tool included as well as the correct bios. You will also need a copy of the same version at the root of the C drive on most systems. The one thing you can try first is a startup floppy already prepared for this. When in the drive and the system is restarted press the F8 key ro bring up the boot menu. Simply choose the "removable" or "floppy" option to test this.

Do you mean when the flash fails or something and it asks for the image from a disk? Because otherwise it's just your basic boot process. Probably just boot to DOS and load the software.

You probably have the EZ-Bios being that the board there is a newer Asus model. The user's manual or the Asus support site is the best place to look for instruction on reflashing the bios with an update. On many of the newer boards you can flash an update on without the need for a boot floppy and the "awdflash" utility for this. Simply make sure the update is at the root of C and enter the onboard flash method according to the manual's instructions.
