What's the main difference between the PCI Video Card and the PCI Express?
I have both and planning to completely rebuild another computer for my
business.Does the new PCI Express card have a bigger data bus, etc.

Thanks for any information

PCI is a 32 bit architecture bus and pci-e is a 64bit architecture. PCI is at end of life right now and pci-e is taking over. Hope that answers your question.

PCI runs on a Parallel bus and can only pass info. on way at a time

PCIe runs on a serial bus and can pass info both ways at the same time and the bandwidth is not shared.

For graphics, PCIe is the best (fastest) graphics interface, below that you have agp (wich is getting obsolete and expensive), below that you have PCI (just plain old an slow). Usally PCI graphics cards are an alternative to onboard graphics card for non gaming PCs/simple work computers, or are also used to plug in more monitors than the main graphics card can.
